Two different questions, hiding in one search

"How long does it take to work" usually means one of two different things: how fast does the magnesium get absorbed, or how long before your overall magnesium status changes. The research answers these on very different timelines, and mixing them up is where a lot of marketing gets loose.

What absorption trials measured: hours

A 1994 crossover trial gave 12 patients a single 100 mg dose of magnesium diglycinate (labeled with a magnesium isotope to track it precisely) and sampled blood and urine over the following 24 hours. Peak isotope enrichment appeared a mean of 3.2 hours earlier after the glycinate dose than after an oxide dose, and absorption was measurable well within that same day. That trial answers the "how fast does it get absorbed" question: in hours, not days.

What status-repletion trials measured: weeks

A 2003 trial gave 46 healthy adults 300 mg of elemental magnesium (as amino-acid chelate, citrate, or oxide) once daily and measured blood, urine and saliva magnesium twice: 24 hours after the first dose ("acute"), and again after 60 days of daily dosing ("chronic"). The organic forms showed greater urinary magnesium excretion at 60 days than at the start, and blood and saliva differences between forms only became clear at the 60-day mark, not the 24-hour one. That's the timeline for a measurable change in overall magnesium status: weeks of consistent daily intake, not a single dose.

Why we don't give a "you'll feel it in X days" answer

Neither trial above measured a felt effect — mood, energy, sleep, or anything else you'd notice day to day. They measured magnesium concentrations in blood, urine and saliva.
