Research review
Does berberine help with sleep? What the research does and doesn't show
No human trial has tested berberine on its own for sleep. What exists is a rat study and a single insomnia report from a small human trial of a multi-ingredient supplement.
No human trial has tested berberine, on its own, as a sleep aid. What exists is one rat study on an insomnia model, and a single reported case of insomnia in a small human trial of a multi-ingredient supplement that included berberine alongside three other compounds — not evidence isolating berberine's effect on sleep either way.
What the evidence doesn't support
It doesn't support berberine as a sleep aid, a treatment for insomnia, or something that improves sleep quality. No human trial has given people berberine on its own and measured sleep as an outcome. It also doesn't clearly support the idea that berberine causes insomnia — the one relevant human data point comes from a multi-ingredient product, not berberine alone, and involved a single case among many participants.
What the trials found
A 2020 study induced insomnia-like sleep disruption in rats using caffeine and a serotonin-depleting compound, then gave the rats oral berberine daily for seven days. Compared with untreated insomnia-model rats, body weight, memory and sleep-quality measures improved. The study also treated a separate group of rats with the anti-anxiety medication diazepam as a comparator arm; we're not reporting how berberine's results compared to it, since a single rat study isn't a basis for that kind of claim regardless of the outcome. The exact berberine dose used wasn't stated in the published abstract.
Separately, a 2017 randomized, placebo-controlled human trial tested a food supplement combining berberine with red yeast rice, coenzyme Q10 and hydroxytyrosol in 158 people with mild-to-moderate high cholesterol, over four weeks. Five adverse events were reported across the whole trial; one was insomnia, in a participant taking the berberine-containing supplement, judged by investigators to be related to treatment. That's a single case in a product containing three other ingredients besides berberine — not evidence that berberine itself caused it, but it's the only human data point connecting the two that we found.
Questions people ask
Does berberine cause insomnia?
The only human data point is a single case of insomnia reported in a small trial of a four-ingredient supplement that included berberine — one participant out of 158, judged treatment-related. Because three other ingredients were in that same product, it isn't clean evidence that berberine specifically caused it, and no trial has tested berberine on its own for this.
Does berberine help with sleep?
No human trial has tested that. The only supportive finding is a rat study using a chemically induced insomnia model, not a measurement of sleep in a person.
When to talk to a doctor
Insomnia that persists, or sleep problems that affect daily function, are worth raising with a doctor — there are treatments with actual human sleep-outcome evidence behind them. Berberine is not a treatment for insomnia or a sleep aid, and nothing in the evidence above supports using it for that purpose. If new sleep problems start after beginning any supplement, mention it to a doctor, and stop the supplement if the problems don't resolve.
